The cyclic AMP receptor protein (CRP) is a global transcriptional regulator that mediates cellular responses to cyclic adenosine monophosphate (cAMP). Binding of cAMP induces a conformational change in CRP that enables it to bind specific regulatory DNA sequences and modulate transcription of numerous genes. The cAMP-CRP regulatory system is involved in the control of carbohydrate uptake and metabolism and other cellular processes, allowing the bacterium to adjust gene expression in response to nutrient and environmental conditions.
EN
The crp gene is one of the two genes modified to attenuate the Salmonella enterica subsp. enterica serovar Typhimurium strain used in the live vaccine AviPro® Megan® Vac 1; the other modified gene is cyaA, which encodes adenylate cyclase and is responsible for the synthesis of cAMP.
Disruption of crp, together with disruption of cyaA, interferes with the cAMP-CRP regulatory system and contributes to attenuation of the vaccine strain. Experimental studies of S. Typhimurium Δcya Δcrp mutants have shown substantially reduced virulence while retaining the capacity to colonize host tissues and induce protective immune responses. In chickens, Δcya Δcrp mutants were shown to be avirulent while retaining the ability to colonize different tissues, although at lower levels than the wild-type strain.
Whole-genome sequence analysis of AviPro® Megan® Vac 1 confirmed disruption of the crp gene, corresponding to locus STM3466 in the S. Typhimurium LT2 reference genome.
